8-OH-dG (8-hydroxy-2-deoxy Guanosine) 是DNA受活性氧及活性氮等作用造成氧化损伤的产物,是氧化应激的标记物。鸟苷的羟基化可发生于正常的新陈代谢进程或者受外界环境因素影响时。8-OH-dG的增多通常还与一些疾病有关,例如癌症,糖尿病,高血压等。
在一些复杂的样品中,例如血浆,细胞裂解液,以及组织中,8-OH-dG既可以以游离的形式存在也可以以整合于DNA中的形式存在。一旦血液进入肾脏当中,游离的8-OH-dG就会渗出到尿液中,而大分子的DNA碎片则留在血流里。由于血浆样品的复杂性,尿液是更好的用来检测游离8-OH-dG的样品。尿液样品中的8-OH-dG水平大概是2.7~13 ng/mg creatine,而血浆中的游离8-OH-dG水平是4~21 pg/ml。
